Spring barley TL-GP-PIL1 with a mutation in the HvPIL1 (HvPIL1 -2)
EN
HvPIL1 -2
Non
The spring barley (Hordeum vulgare L.) cultivar Golden Promise was modified using CRISPR/Cas-mediated genome editing to generate the HvPIL1 -2 line with reduced plant height. The modification targets the endogenous HvPIL1 (phytochrome interacting factor-like 1) gene, which is involved in the regulation of stem growth. Homozygous HvPIL1 -2 plants exhibited a reduction in height of approximately 10 cm under greenhouse conditions. The reduced stature may contribute to decreased susceptibility to lodging.

Information on the modified DNA sequences
The HvPIL1 -2 line contains a two-base-pair deletion of adenine and cytosine that disrupts the reading frame, resulting in a non-functional HvPIL1 protein. HvPIL1 encodes a basic helix-loop-helix (bHLH) transcription factor involved in the regulation of stem growth. The gene is light responsive and contributes to the activation of genes involved in cell wall expansion. Its expression fluctuates throughout the day, peaking around midday, and is downregulated under drought conditions. Homozygous HvPIL1 -2 mutant plants exhibited a reduction in height of approximately 10 cm when grown under greenhouse conditions. The resulting reduction in plant height is expected to reduce susceptibility to lodging.

Note
  •  Molecular characterization confirmed the intended edit in the HvPIL1 gene and the absence of transgenic sequences derived from the genome-editing vector in the final selected generation.
  •  The precise position of the adenine insertion within the HvPIL1 coding sequence and the resulting altered nucleotide and amino acid sequences have not been provided in the information available for this record.

Caractéristiques de l’OVM
As a light-responsive bHLH transcription factor, HvPIL1 regulates downstream gene expression associated with plant growth and development. In particular, HvPIL1 has been reported to activate genes involved in cell wall expansion and to participate in the regulation of stem growth. Disruption of HvPIL1 function in the HvPIL1 -2 line is therefore expected to alter the transcriptional regulation of downstream genes involved in these processes, consistent with the reduced plant height observed in homozygous mutant plants. Specific downstream target genes and detailed transcriptomic effects of the HvPIL1 -2 mutation have not been provided in the information available for this record.
